Detailed introduction:

Vitamins are compounds that you must have for growth and health. They are needed in
small amounts only and are available in the foods that you eat. Vitamin D is necessary for
strong bones and teeth.
Lack of vitamin D may lead to a condition called rickets, especially in children, in which
bones and teeth are weak. In adults it may cause a condition called osteomalacia, in
which calcium is lost from bones so that they become weak. Your doctor may treat these
problems by prescribing vitamin D for you. Vitamin D is also sometimes used to treat other
diseases in which calcium is not used properly by the body.
Alfacalcidol is a form of vitamin D used to treat hypocalcemia (not enough calcium in the
blood). Alfacalcidol is also used to treat certain types of bone disease that may occur with
kidney disease in patients who are undergoing kidney dialysis. 
Alfacalcidol can facilitate calcium reabsorption at small intestine and renal tubule. It helps
inhibiting parathyroid hyperplasia and related hormone synthesis and releasing. 
Alfacalcidol also promotes collagen and bone matrix protein synthesis by increase
synthesis of TGF-¦Â and IGF-I.
Alfacalcidol regulates muscular calcium metabolism and facilitates muscle cell
differentiation to strengthen muscle power.
Pharmacokinetics
Faneng is absorbed in small intestines after oral administration, then is converted into
1,25-(OH)2D3 via hepatic metabolism. Now it is known that osteoblasts express mRNA of
25-hydroxylase which can convert 1¦Á-OH-D3 into active form. The Cmax of active
1¦Á-OH-D3 appears at 8~12 hrs after administration with half life of 17.6 hrs.
Indications
Faneng is indicated for:
1. Rickets and osteomalacia.
2. Renal osteodystrophy.
3. Osteoporosis.
4. Hypoparathyroidism.
Adverse Reactions
Faneng administered alone with small dose (<1.0¦Ìg/d) usually has no obvious adverse
reactions. Long term usage with high dose or combine with calcium agents may cause
hypercalcemia and hypercalciuria.
Precautions
1. Regular tests of plasma Calcium, blood urea nitrogen, creatinine, and urinary calcium
and creatinine in urine during usage with the doctor's advices is recommended.
2. Faneng is only indicated for use in young patients with youth idiopathic osteoporosis
and excessive corticosteroid induced osteoporosis.
3. It's necessary to withdraw this drug when hypercalcemia occurs and go to the doctor for
appropriate treatment. Half dose of last dosage should be the initial dose when restart
medication after blood calcium recovered.
Dosage and Administration
Oral administration. 
For adults: the daily therapeutic dose is 0.25~1¦Ìg, daily maintenance dose is 0.25¦Ìg.
For pediatric use: 0.25¦Ìg every two days or according to prescription.
